Sorry to get your hopes up but it's not possible from a script (without a large amount of work and changes to the original code). Whilst it is possible to change the setting on a key press, the UI checkbox would not automatically update. So, the check box would not reflect the actual status of select-through.

Perhaps consider leaving select-through ticked and using shift+control+left-mouse to 'drag-select' the nearest geometry. (use shift+control+right-mouse to remove from selection)
